The primary amyloid-beta analysis compares CSF amyloid-beta levels at month 3 and month 6 with paired-sample t-tests. The primary outcomes are morning CSF amyloid-beta 40 and amyloid-beta 42 levels before and after treatment. Additional biomarkers include plasma amyloid-beta and CSF amyloid-beta 38, amyloid-beta 43, total tau, and phosphorylated tau 181. Participants without required lumbar punctures are excluded from the primary outcome analysis. The alpha threshold for amyloid-beta testing is Bonferroni-corrected to 0.025 because two amyloid-beta parameters are tested. The trial collects primary CSF biomarkers by lumbar puncture at month 3 and month 6.
