Each CAPOX course consists of oxaliplatin 130 mg/m2 by 2-hour intravenous infusion on day 1, plus oral capecitabine from the evening of day 1 through the morning of day 15, on a 21-day cycle. All trial participants receive 6 months of CAPOX adjuvant chemotherapy for up to 8 courses. The capecitabine starting dose is reduced to 1500 mg/m2/day for patients with creatinine clearance between 30 and 50 mL/min or aged 70 years or older. A prior large Japanese trial of postoperative CAPOX adjuvant chemotherapy reported chronic OIPN of CTCAE grade 2 or higher in approximately 36% of patients.
