Participants assigned to orforglipron began at 12 mg once daily and escalated every four weeks to 36 mg or their maximum tolerated dose. Orforglipron is a once-daily nonpeptide oral GLP-1 receptor agonist being investigated for obesity and type 2 diabetes. Orforglipron can be taken without restrictions on food or water intake. Direct transition to 12 mg orforglipron after injectable incretin therapy was generally well tolerated.
