CAAR T cells use self-antigen display to kill autoreactive B cells while preserving the rest of the B-cell repertoire. IGHV4-34-targeted CAR T cells were developed to selectively eliminate lupus-associated B-cell clones while sparing normal B cells. T-cell-driven diseases may be treated by selectively depleting pathogenic T-cell clones. Pan-B-cell depletion also removes protective humoral immunity, motivating more selective approaches.
