Pretomanid showed the highest signals among the three drugs for supraventricular extrasystoles and acute cardiac failure. The mechanism of frank pretomanid-associated acute kidney injury in MDR-TB patients remains unknown. Pretomanid's adverse event profile includes gallbladder and pancreatic signals in addition to shared cardiac and hepatic signals. Pretomanid unique signals included cholecystitis, pancreatitis, leukocytosis, acute kidney injury, and multiple organ dysfunction syndrome. Acute kidney injury was detected exclusively for pretomanid and is not listed in any approved label for the three drugs.
