By Clinical Disease Activity Index, low disease activity increased from 13% at baseline to 75% at month 24 among patients still receiving tofacitinib. Drug survival was similar between tofacitinib monotherapy and tofacitinib plus MTX. Real-world tofacitinib drug survival was lower than estimates from long-term extension studies of randomized trials because those enrolled patients pre-selected for acceptable efficacy and tolerability. Tofacitinib is a Janus kinase inhibitor with efficacy and safety demonstrated in randomized controlled trials as both monotherapy and in combination with methotrexate.
